As part of her/his work, the molecular biology engineer is the dedicated molecular biology operator of a client’s project. He/she will be involved in the project analysis, the design and ordering of optimized nucleotide transgene sequences, the preparation of the corresponding expression vectors, as well as in quality control analyses: verification of a transgene integrated into a cell line by PCR amplification from genomic DNA and amplicon sequencing (Sanger and NGS), mycoplasma testing and assessment of the number of copies integrated in cell lines by ddPCR. The writing of reports as well as the associated documentation are also an important part of his/her task. The implementation of new methods and technologies, as well as the participation in R&D projects constitute another aspect of this position. 4. General requirements Conduct clients project through all the molecular biology steps of cell line development at Selexis. This includes the preparations of DNA and multiple analysis in the lab, as well as in-silico analysis and report writing. Sequences optimization including signal peptide screening using bioinformatics tools Plasmid cloning and preparation of expression vectors in E. coli Purification of DNA fragments for electroporation Genomic DNA extraction and cDNA preparation PCR and purification of PCR products for Sanger Sequencing PCR and preparation of the plate for NGS Sequencing Sanger and NGS Sequencing analysis qPCR (TaqMan and SYBER Green) ddPCR (copy number analysis) Mycoplasma testing by PCR Writing of SOPs and Forms Writing of the documentation and analysis reports Implementation of new methods / technologies in the molecular biology lab Carry out or participate in R&D projects Internal project presentation 5.
